摘要:
目的 探讨酪氨酸激酶受体1短发夹RNA（Tie1-shRNA）对高糖培养的大鼠内皮细胞（rECs）Tie1的表达的影响。 方法 将rECs分为对照组、高糖组、高糖+慢病毒组和高糖+Tie1-shRNA组4组。构建Tie1-shRNA慢病毒载体，作用于高糖培养的rECs，干扰酪氨酸激酶受体1（Tie1）mRNA的表达；采用MTT法检测细胞增殖情况，细胞划痕实验检测细胞迁移情况，Western blot检测rECs中Tie1、血管内皮生长因子-A（VEGF-A）、基质金属蛋白酶-2（MMP-2）和基质金属蛋白酶-9（MMP-9）的蛋白表达。 结果 细胞增殖活性4组间差异有统计学意义（F=48.937，P<0.01），其中高糖组细胞增殖活性明显高于对照组（P<0.01），高糖+Tie1-shRNA组细胞增殖活性较高糖组和高糖+慢病毒组均明显减弱（均P<0.01）。细胞迁移率4组间差异有统计学意义（F=26.728，P<0.01），其中高糖组细胞迁移率比对照组明显增加（P<0.01），高糖+Tie1-shRNA组细胞迁移率较高糖组和高糖+慢病毒组均明显减弱（均P<0.05）。4组rECs的Tie1、VEGF-A、MMP-2、MMP-9蛋白的表达差异有统计学意义（FTie1=85.902，P<0.01；FVEGF-A=48.937， P<0.01；FMMP-2=169.857，P<0.01；FMMP-9=130.018，P<0.01），其中高糖组rECs的Tie1、VEGF-A、MMP-2、MMP-9蛋白的表达明显高于对照组（均P<0.05），高糖+Tie1-shRNA组rECs的Tie1、VEGF-A、MMP-2、MMP-9蛋白的表达较高糖组和高糖+慢病毒组均明显降低（均P<0.05）。 结论 通过Tie1-shRNA技术抑制高糖培养的rEC中Tie1的表达，可下调VEGF-A、MMP-2、MMP-9蛋白的表达，减轻高糖诱导的rEC的增殖和迁移。
Abstract:
Objective To investigate the effect of Tie1-shRNA on the expression of tie1 in high glucose cultured rat endothelial cell（rECs）．Methods rECs were divided into four groups：control group, high glucose group, high glucose + lentivirus group and high glucose + Tie1-shRNA group．Tie1-shRNA lentiviral vector was constructed and used to interfere the expression of Tie1 mRNA in high glucose cultured rECs．The viability of rEC was detected by MTT, the migration of rEC was detected by cell scratch tests, and the expressions of Tie1, VEGF-A, MMP2 and MMP-9 were detected by Western blot．Results The difference of cell viability among the four groups was statistically significant (F=48.937, P<0.01).The cell viability of high glucose group was significantly higher than that of control group (P<0.01)．The cell viability of high glucose + Tie1-shRNA group was lower than that of high glucose group and high glucose + lentivirus group (both P<0.01)．The difference of cell migration rate among the four groups was statistically significant (F=26.728, P<0.01)．The cell migration rate of high glucose group was significantly higher than that of control group (P<0.01), and the cell migration rate of high glucose + Tie1-shRNA group was significantly lower than that of high glucose group and high glucose + lentivirus group (both P<0.05)．The differences of the expression of Tie1, VEGF-A, MMP-2 and MMP-9 protein among the four groups were statistically significant (FTie1=85.902, P<0.01; FVEGF-A=48.937, P<0.01; FMMP-2=169.857, P<0.01; FMMP-9=130.018, P<0.01)．The expression of Tie1, VEGF-A, MMP-2 and MMP-9 protein of high glucose group were significantly higher than those of control group (all P<0.05).The expression of Tie1, VEGF-A, MMP-2 and MMP-9 protein in high glucose + Tie1-shRNA group were significantly lower than those in high glucose + lentivirus group and high glucose group (all P<0.05)．Conclusion Tie1-shRNA can decrease the expression of Tie1, VEGF-A, MMP-2 and MMP-9, suppress cell viability and cell migration induced by high glucose.
