摘要:
目的 通过生物信息学的方法分析炭疽毒素受体1(ANTXR1)在胃癌中表达及意义,揭示相应的分子机制。方法 检索多个公共数据库中有关ANTXR1表达和临床数据,分析ANTXR1 在胃癌中表达情况及其与包括预后在内的临床病理特征的关系,并下载ANTXR1的共表达基因,通过基因本体(GO)富集和京都基因与基因组百科全书(KEGG)通路分析预测ANTXR1参与的生物学过程。结果 ANTXR1 在胃癌组织中的表达异常上调,其表达与胃癌患者的T 分期、肿瘤分期、组织分化以及种族相关; 并且ANTXR1高表达的患者预后差(P均<0.05)。ANTXR1共表达基因主要参与细胞外基质组成、细胞外结构、胚胎器官发育等过程,并显著富集于多个致癌信号通路、细胞黏附分子、ECM-受体相互作用等相关通路。结论 ANTXR1高表达与胃癌患者预后不良有关,在胃癌发生发展中发挥着促癌作用,可能是胃癌的一个可靠的诊疗靶点。
Abstract:
Objective To investigate the expression and significance of ANTXR1 in gastric cancer(GC), and to explore the potential molecular mechanisms. Methods We retrieved the expression and clinical data of ANTXR1 in several public databases, and analyzed the expression of ANTXR1 in GC and its relationship with clinicopathological features including prognosis. We further downloaded the co-expression gene of ANTXR1 and predicted the biological processes involved in ANTXR1 by GO and KEGG analysis. Results Compared to normal tissues, ANTXR1 was significantly upregulated in GC. The expression of ANTXR1 was significantly correlated with T stage, tumor stage, tissue differentiation and race of gastric cancer patients. The prognosis of patients with high ANTXR1 expression was poor. All the above analyses had statistical significance(P<0.05). The co-expression genes of ANTXR1 were mainly involved in extracellular matrix composition, extracellular structure, embryonic organ development. Moreover, these genes were significantly enriched in several carcinogenic signaling pathways, cell adhesion molecules and ECM-receptor interactions. Conclusion The high expression of ANTXR1 is associated with poor prognosis of GC patients, which plays a role in promoting the development of GC. It may be a reliable target for diagnosis and treatment of GC.
