摘要:
目的 探讨microRNA-365(miR-365)对骨肉瘤大鼠巨噬细胞增殖和凋亡的影响。方法 构建骨肉瘤大鼠模型,采用右后肢外侧皮下注射的方法将miR-365模拟物、miR-365阴性对照物、miR-365 抑制物、miR-365 抑制阴性对照物及生理盐水注入到大鼠模型中,采用RT-PCR检测骨肉瘤大鼠巨噬细胞miR-365、KRAS的mRNA表达,Western blot检测骨肉瘤大鼠巨噬细胞周期相关蛋白cyclin D1 和 cyclin E的表达。结果 与miR-365阴性对照组、miR-365抑制组、miR-365抑制阴性对照组、空白组相比较,miR-365组的miR-365、KRAS的mRNA表达量及cyclin D1、cyclin E的蛋白表达升高,差异性有统计学意义(P<0.05); 与miR-365抑制阴性对照组、空白组相比较,miR-365抑制组的miR-365、KRAS的mRNA表达量及cyclin D1、cyclin E的蛋白表达降低,差异有统计学意义(P<0.05); miR-365阴性对照组、miR-365抑制阴性对照组、空白组之间miR-365、KRAS的mRNA表达量及cyclin D1、cyclin E的蛋白表达差异无统计学意义(P>0.05)。结论 miR-365能够通过KRAS促进巨噬细胞的增殖,减轻生物体炎症反应。
Abstract:
Objective To investigate the effects of microRNA-365 on macrophage proliferation and apoptosis in osteosarcoma rats.Methods The osteosarcoma rats model was established.miR-365 simulant,miR-365 negative control,miR-365 inhibitor,miR-365 inhibitor negative control and normal saline were injected into the rat model.The mRNA expression of miR-365 and KRAS in macrophages of osteosarcoma rats was detected by RT-PCR, and the expression of cyclin D1 and cyclin E were detected by Western blot.Results Compared with miR-365 negative control group, miR-365 inhibition group, miR-365 inhibition negative control group and blank group, the mRNA expression levels of miR-365 and KRAS and the protein expression levels of cyclin D1 and cyclin E in miR-365 group were increased, and the differences were statistically significant(P<0.05).Compared with miR-365 inhibition negative control group and blank group, the mRNA expression of miR-365and KRAS and the protein expression of cyclin D1 and cyclin E in miR-365 inhibition group weredecreased,and the differences were statistically significant(P<0.05).There were no significant differences in the mRNA expression of miR-365 and KRAS and the protein expression of cyclin D1 and cyclin E between miR-365 negative control group, miR-365 inhibition negative control group and blank group(P>0.05).Conclusion miR-365 can promote the proliferation of macrophages through KRAS and reduce the inflammatory reaction of organism.
