摘要:
目的 探讨衣康酸二甲酯对非肥胖性糖尿病小鼠(NOD)胰岛炎的影响。方法 利用流式细胞仪检测衣康酸二甲酯对RAW 264.7巨噬细胞表型的影响,通过ELISA观察其对RAW 264.7分泌的细胞因子(IL-6、IL-12和TNF-a)的影响,同时将衣康酸二甲酯腹腔注射NOD小鼠,进一步探讨其对胰岛内部炎症的影响。结果 衣康酸二甲酯有利于降低巨噬细胞M1/M2比率,使巨噬细胞向M2型转换,同时降低IL-6、IL-12和TNF-α的分泌,50 mg/kg的衣康酸二甲酯使胰岛评分达到了1.115±0.782 1。结论 衣康酸二甲酯通过M1/M2巨噬细胞表型转化可以缓解NOD小鼠胰岛炎。
Abstract:
Objective To investigate the effect of dimethyl itaconate on insulitis in NOD mice.Methods The phenotypic changes of RAW 264.7 macrophages induced by dimethyl itaconate were detected by flow cytometry, and the effect of dimethyl itaconate on the secretion of RAW 264.7 cytokines(IL-6、IL-12 and TNF-α)were observed by ELISA.At the same time, dimethyl itaconate was injected intraperitoneally into NOD mice to further explore its effect on islet inflammation.Results Dimethyl itaconate reduced the M1/M2 ratio of macrophages and converted macrophages to M2 type, and also reduced the secretion of IL-6, IL-12 and TNF-α.The islet score of 50 mg/kg dimethyl itaconate reached 1.115±0.782 1.Conclusion Dimethyl itaconate can alleviate insulitis in NOD mice by phenotypic transformation of M1/M2 macrophages.
