摘要:
目的 研究低剂量三唑磷暴露、高糖高脂饮食对二乙基亚硝胺(DEN)诱发的大鼠肝癌模型肝组织病理的影响。方法 将正常6周龄雄性SD大鼠70只随机分为4组,分别为对照组(10只)、DEN组(20只)、农药组(20只)、双高组(20只)。除对照组外,其余3组分别进行间歇腹腔注射DEN、腹腔注射DEN+有机磷农药饮水、腹腔注射DEN+高脂高糖饲料的处理,观察大鼠的饮食、毛色、精神及活动情况,每周测量大鼠体质量,每隔4周解剖部分大鼠观察肝脏形态,造模16周时解剖大鼠观察肝脏病理学形态。结果 对照组大鼠饮食活动正常,毛色自然。实验组随着DEN注射次数的增加,均出现不同程度的消瘦、脱毛、毛发枯槁、活动减少等现象。造模后,各实验组大鼠平均体质量均低于对照组。实验第16周时,DEN组、农药组及双高组的肝脏形态皆出现明显的病理改变,各组的病变严重程度为:双高组>农药组>DEN组。 结论 农药污染、高糖高脂饮食可促进DEN诱导肝脏由肝炎-肝硬化的发展过程; 施加低剂量三唑磷、高糖高脂饮食因素诱导的肝癌发展过程在组织形态学上更接近临床真实病例,这为改进DEN肝癌大鼠模型提供有力的实验依据。
Abstract:
Objective To study the effects of low-dose triazophos exposure, high-sugar and high-fat diet on the pathological changes of liver tissue in rat liver cancer models induced by diethylnitrosamine(DEN).Methods A total of 70 normal male SD rats,6-week-old,were randomly divided into 4 groups, namely the control group(n=10), the DEN group(n=20), the pesticide group(n=20), and the Shuang Gao group(n=20).Except for the control group, the other 3 groups were treated with intermittent hypodermic injection of DEN, hypodermic injection of DEN plus drinking water added with triazophos, and hypodermic injection of DEN plus high-fat and high-sugar feed.The diet, hair color, and activity of rats were observed weekly.The body weight of the rats was measured every week.Some rats were dissected to observe liver morphology every 4 weeks.HE staining was used to obtain the liver pathology at week 16. Results The rats in the control group had normal eating activities and natural hair color.With increasing number of DEN injections in the experimental groups, there were various degrees of weight loss, hair loss, atrophy, and decreased activity.After modeling, the average weight of rats in each experimental group was lower than that of the control group.At the 16th week of the experiment, the liver morphology of the DEN group, the pesticide group and the Shuang Gao group showed obvious pathological changes.The severity of liver fibrosis in each group was: Shuang Gao grouP>pesticide grouP>DEN group.Conclusion Low-dose triazophos exposure, high-sugar and high-fat diet can promote the development of DEN-induced liver cancer.Histological morphology of the liver cancer model induced by low-dose triazophos, high-sugar and high-fat diet is closer to that of clinical liver cancer cases, which provides powerful experimental basis for improving DEN liver cancer rat model.
