摘要:
目的 探讨血红素加氧酶-1(HO-1)在脊髓损伤(SCI)中的保护作用及其机制。方法 采用压迫损伤法构建SCI模型,将大鼠随机分为假手术组(Sham组)、单纯SCI组(Vehicle组)、腺相关病毒感染SCI大鼠组(A-C组)和过表达HO-1的腺相关病毒感染SCI大鼠组(A-HO-l组)。Western blot检测HO-1、NLRP1、NLRP3、NLRC4和caspase-1的表达,免疫荧光检测HO-1和MAP2的表达定位,原位缺口末端标记法(TUNEL)检测脊髓组织细胞凋亡,后肢行为学(Basso-Beattie-Bresnahan,BBB)评分评估大鼠后肢运动功能。结果 SCI后HO-1表达增加,差异有统计学意义(P<0.05)。与A-C组相比,A-HO-1组中HO-1水平明显升高,NLRP1、pro-caspase-1、caspase-1中p20表达水平和凋亡细胞数显著降低,差异有统计学意义(P<0.05)。损伤后3、7、14、21 d,A-HO-1组大鼠BBB评分较Vehicle组和A-C组显著升高,差异有统计学意义(P<0.05)。结论 过表达HO-1通过抑制NLRP1炎性复合物激活而减少SCI后细胞凋亡和改善大鼠后肢运动功能,对SCI大鼠起保护作用。
Abstract:
Objective To investigate the protective effect of heme oxygenase-1(HO-1)on spinal cord injury(SCI)and its mechanism.Methods SCI model was established by compression injury, and the rats were randomly divided into sham group, simple SCI group(Vehicle group), adeno-associated virus infected SCI group(A-C group)and adeno-associated virus overexpressing HO-1 infected SCI group(A-HO-l group).The expression of HO-1, NLRP1, NLRP3, NLRC4 and caspase-1 was detected by Western blot, HO-1 and MAP2 expression were detected by immunofluorescence, cell apoptosis of spinal cord tissues was measured by TUNEL, and motor function of hind limbs was evaluated by Basso-Beattie-Bresnahan(BBB)score.Results The expression of HO-1 was increased significantly after SCI(P<0.05).Compared with A-C group, the expression of HO-1 increased significantly, the expression of NLRP1、pro-caspase-1、caspase-1 p20 and the proportion of apoptotic cells decreased significantly in A-HO-1(P<0.05)group.The BBB score was significantly higher in the A-HO-1 group than that in the Vehicle and A-C groups at 3, 7, 14, and 21 days after SCI(P<0.05).Conclusion Overexpression of HO-1 can protect SCI rats by inhibiting the activation of NLRP1 inflammasome, reducing cell apoptosis and improving motor function of hind limbs.
