摘要:
目的 探讨肿瘤相关成纤维细胞(CAF)通过FGFR4/FGF19信号轴调控结直肠癌生长可能机制。方法 将研究对象分为3组,分别是细胞组、细胞+NF组、细胞+CAF组。绘制各组癌细胞生长曲线,采用ELISA法检测细胞外FGF19因子浓度变化。 结果 癌细胞与CAF共培养后,癌细胞生长率明显提高,与其他组比较,差异有统计学意义(P<0.05); 细胞外FGF19因子浓度明显提高,与其他组比较,差异有统计学意义(P<0.05)。结论 CAF通过促进结肠癌细胞自分泌FGF19因子,促进癌细胞分裂生长。
Abstract:
Objective To explore the possible mechanism of cancer associated fibroblast(CAF)regulating the growth of colorectal cancer through the signal axis of fibroblast growth factor 4/fibroblast growth factor 19. Methods The experiment was divided into three groups, including cancer cell group, cancer cell+NF group, cancer cell+CAF group. The growth curve of each group was drawn. The changes of extracellular fibroblast growth factor 19 were detected by ELISA. Results After co-culture of cancer cells with CAF, the growth rate of cancer cells was significantly increased, compared with other groups(P<0.05); the concentration of extracellular fibroblast growth factor 19 was significantly increased, compared with other groups(P<0.05). Conclusion CAF can promote the autocrine of fibroblast growth factor 19 and promote the growth of cancer cells.
