目的 基于生物信息学方法筛选骨肉瘤(OS)的关键基因及功能预测。方法 检索GEO 数据库OS数据集(GSE14359),使用GEO2R筛选差异基因(DEGs),构建DEGs的蛋白质-蛋白质相互作用网络(PPI)以及Cytoscape可视化分析确定关键基因。并进一步下载关键基因共表达基因,基因本体(GO)和(京都基因与基因组百科全书)KEGG分析预测其可能的生物功能。结果 通过对原发OS组织、OS肺转移组织和原代成骨细胞之间的差异性和维恩图分析,获得385个交集基因。进而使用蛋白质-蛋白质相互作用(PPI)和 Cytoscape 从交集基因中筛选出关键基因GMFG。 GMFG共表达基因主要参与中性粒细胞激活、中性粒细相关的免疫反应等过程,并显著富集于核糖体细胞处理过程、白细胞跨膜转运过程等通路。结论 GMFG作为OS关键基因,在OS进展中发挥促癌作用,可能是OS的可靠的诊疗靶点。
Abstract:
Objective To screen the key genes and functional prediction of osteosarcoma(OS)based on bioinformatics.Methods The OS dataset(GSE14359)from GEO database were retrieved, and GEO2R to screen the differential genes(DEGs)were used.We further constructed the protein and protein interaction network(PPI)of DEG, and identified the key gene through the visualized analysis of Cytoscape.The coexpression genes of key gene were downloaded, and the possible biological function was predicted by GO and KEGG analysis.Results By difference analysis and Venn diagram between conventional osteosarcoma tissue, osteosarcoma lung metastasis tissue and non-neoplastic primary osteoblast cells, we obtained 385 intersection genes.Then, PPI and Cytoscape were used to screen the key gene(GMFG).The co-expression genes of GMFG were mainly involved in neutrophil activation, neutrophil activation involved in immune response, and were significantly enriched in ribosome cell processing, leu-kocyte transendothelial migration.Conclusion As a key gene of OS, GMFG plays a role in promoting carcinogenesis in the progression of OS, which may be a reliable diagnosis and treatment target of OS.
