摘要:
目的 探讨SPOCK1基因在泛癌中的诊断价值及肿瘤预后评估的作用。方法 通过基因型组织表达(GTEx)、癌症细胞系百科全书(CCLE)及癌症基因组图谱(TCGA)在内的多个数据库,评估SPOCK1基因在泛癌分析中的表达模式,ROC曲线分析SPOCK1基因在泛癌中的诊断价值,Cox回归及Kaplan-Meier分析在泛癌中SPOCK1基因表达与肿瘤预后的相关性。结果 多个数据库的差异分析结果显示SPOCK1的表达在泛癌中不一致的上调或下调,表达模式存在内在的差异。ROC曲线显示SPOCK1在预测14种癌症类型方面具有一定的准确性(AUC>0.7),在预测宫颈鳞癌和腺癌、胆管癌和睾丸癌具有较高的准确性(AUC>0.9)。生存分析则提示SPOCK1高表达与膀胱尿路上皮癌、乳腺浸润癌、肾透明细胞癌、肺腺癌、胰腺癌、嗜铬细胞瘤、副神经节瘤、间皮瘤及前列腺癌的低存活率相关。结论 SPOCK1可能是泛癌诊断和预后的重要分子生物标志物。
Abstract:
Objective To investigate the diagnostic value and prognosis of SPOCK1 gene in pan-cancer.Methods Through multiple databases including Genotype-Tissue Expression, Cancer Cell Line Encyclopedia and the Cancer Genome Atlas, the expression pattern and the diagnostic value of SPOCK1 gene in pan-cancer was evaluated, and the correlation between SPOCK1 gene expression and tumor prognosis was analyzed by Cox regression and Kaplan Meier.Results The difference analysis results of multiple databases showed that the expression of SPOCK1 was up-regulated or down-regulated inconsistently in pan-cancer, and there were internal differences in expression patterns.ROC curve showed that SPOCK1 had certain accuracy in predicting 14 cancer types(AUC>0.7), and had high accuracy in predicting cervical squamous cell carcinoma, adenocarcinoma, cholangiocarcinoma and testicular cancer(AUC > 0.9).Survival analysis showed that the high expression of SPOCK1 was associated with low survival rate in bladder urothelial carcinoma, invasive breast cancer, clear cell renal cell carcinoma, lung adenocarcinoma, pancreatic cancer, pheochromocytoma, paraganglioma, mesothelioma and prostate cancer.Conclusion SPOCK1 may be an important molecular biomarker for diagnosis and prognosis of pan-cancer.
