目的 通过将血糖仪和血管内皮生长因子(VEGF)适配体传感器相结合,构建一种便携式适配体传感系统,并将其用于骨肉瘤标志物的检测。方法 将血糖仪作为检测器,与适配体传感器相结合,开发出一种便携式血糖仪联合VEGF适配体传感系统,在对其反应条件(孵育时间、反应时间、存储时间)进行优化的基础上测定了其检测限。同时,选取10份骨肿瘤患者血清进行检测,通过与传统的ELISA法进行比较,评估其准确度。结果 成功构建了一种便携式血糖仪联合VEGF适配体传感系统,其最佳孵育时间60 min、反应时间30 min、存储时间7 h,最低检测限为6.5×10-9mol/L(2.58 μg/kg)。该法用于骨肉瘤患者血清的检测,与ELISA 法检测结果差异无统计学意义(t=2.322,P>0.05)。结论 成功构建了一种便携式血糖仪联合VEGF适配体传感系统,并将其运用到骨肉瘤患者血清检测,其检测限低,准确性好。
Abstract:
Objective To construct a portable aptamer sensing system by combining a blood glucose meter with a vascular endothelial growth factor(VEGF)aptamer sensor, and use it for the detection of osteosarcoma markers.Methods A portable blood glucose meter combined with a VEGF aptamer sensing system was developed, which was determined on the basis of optimizing its reaction conditions(incubation time, reaction time, storage time).At the same time, ten serum samples from bone tumor patients were selected for testing, and their accuracy was evaluated by comparing with traditional ELISA methods.Results A portable blood glucose meter combined with a VEGF aptamer sensing system was successfully constructed.The optimal incubation time was 60 minutes, the reaction time was 30 minutes, and the storage time was 7 hours.The minimum detection limit was 6.5×10-9mol/L(2.58 μg/kg).This method was used for the detection of serum in patients with osteosarcoma which was not statistically significant difference with ELISA method(t=2.322,P>0.05).Conclusion A portable blood glucose meter combined with a VEGF aptamer sensing system is successfully constructed and applied to detect serum of osteosarcoma patients.Its low detection limits and accuracy was good.
