摘要:
目的 研究Kisspeptin对胰岛β细胞的凋亡影响及可能的作用机制。方法 取16只SD大鼠胰腺组织分为4组:1)对照组:在20%葡萄糖生理盐水中培养; 2)Kp组:在20%葡萄糖生理盐水培养液中加入1 μmol/L Kisspeptin; 3)PD组:在20%葡萄糖生理盐水培养液中加入50 μmol/L ERK1/2信号通路抑制剂PD98059; 4)Kp+PD组:在20%葡萄糖生理盐水培养液中加入1 μmol/L Kisspeptin 和50 μmol/L PD98059。各组在恒温CO2培养箱培养48 h后制作TUNEL染色切片,每组随机取3张切片采用医学图像分析系统IPPS 6.0软件计算每个胰岛组织β细胞凋亡指数,比较每组平均β细胞凋亡指数差异。结果 与对照组 [(4.08±0.42)%]、PD组[(4.10±0.42)%]、Kp+PD组[(4.11±0.47)%] 凋亡指数相比较,Kp组[(3.11±0.46)%]明显下降(P均<0.01); PD组与对照组、Kp+PD组与对照组、Kp+PD组与PD组之间比较的差异均无统计学意义(P>0.05)。结论 Kisspeptin对β细胞凋亡具有抑制作用, ERK1/2信号传导通路可能是其主要的作用通路之一。
Abstract:
Objective To study the effect of Kisspeptin on the apoptosis of islet β cells and its possible mechanism.Methods Pancreatic tissues of 16 SD rats were divided into four groups: 1)control group: cultured in 20% glucose saline; 2)Kp group: 1 μmol/L Kisspeptin was added to 20% glucose saline solutions; 3)PD group: 50 μmol/L PD98059 was added to 20% glucose saline solution; 4): Kp+PD group: 1 μmol/L Kisspeptin and 50 μmol/L PD98059 were added to 20% glucose saline solution.TUNEL staining sections were prepared in a constant temperature CO2 incubator for 48 hours.Three sections were randomly selected from each group.The β-cell apoptosis index of each islet tissue was calculated by the medical image analysis system IPPS 6.0 software and the average β-cell apoptosis index of each group was compared.Results Compared with the control group [(4.08±0.42)%], the PD group[(4.10±0.42)%], and the Kp+PD group [(4.11±0.47)%], the average apoptotic index of the Kp group[(3.11±0.46)%] was significantly decreased(P<0.01 for each).There was no significant difference between PD group and control group, Kp+PD group and control group, Kp+PD group and PD group(P>0.05).Conclusion Kisspeptin has an inhibitory effect on β-cell apoptosis, and ERK1/2 signaling pathway may be one of the main pathways.
