摘要:
目的 研究卵巢癌外泌体的差异表达微小RNA(miRNA)和富集通路,筛选诊治、预后判断分子标志物。 方法 从高通量基因表达数据库(GEO)下载并分析获得在卵巢癌外泌体明显富集差异表达的miRNA基因数据集(GSE)76449。应用生物信息学和蛋白质谱分析方法,筛选外泌体携运miRNA的靶向通路和预后靶标。结果 共筛选出32个外泌体富集差异表达miRNAs,基因本体(GO)和代谢途径分析数据库(KEGG)分析显示,差异miRNAs基因主要富集在细胞氮化合物代谢功能以及鞘糖脂生物合成——乳酸和内酯系列和粘蛋白型O-聚糖生物合成等信号通路,预后分析发现miRNA-337-3p高表达、miRNA-6809-3p预测靶基因WNT3低表达和共表达蛋白卷曲蛋白4基因(FZD4)、干细胞生长因子(HGF)都与卵巢癌不良预后密切有关。结论 外泌体富集葡萄糖代谢相关miRNAs可能与卵巢癌发生发展有关,可为卵巢癌的诊疗提供新思路。
Abstract:
Objective To study differentially expressed miRNAs and enrichment pathways in exosomes derived from ovarian cancer and screen molecular markers for diagnosis, treatment and prognosis.Methods One set of exosomal miRNA expression profile data related to ovarian cancer(GSE76449)were downloaded from GEO database, then up-regulated miRNAs were obtained by differentially expressed analysis.The combination usage of bioinformatics and mass spectrometry were analyzed for functional annotation pathway and prognostic biamarkers.Results A total of 32 differentially expressed miRNAs were obtained.Based on the GO and KEGG function annotation, miRNA were mainly enriched in the function of cellular nitrogen compound metabolic process and the pathway of glycosphingolipid and mucin type O-glycan biosynthesis.By analyzing prognosis of network node, miR-337-3p were found to be upregulated.MiR-6809-3p predicted target gene WNT3 and coexpressed proteins including FZD4、HGF were all associated with the poor outcome of patients with ovarian cancer.Conclusion Exosomal miRNAs influenced glucose metabolism is related to the progress of ovarian cancer and provided new strategies for the diagnosis and treatment of ovarian cancer.
