摘要:
目的研究香加皮杠柳苷(CPP)对小鼠骨髓瘤细胞增殖、凋亡及移植成瘤的影响及作用机制.方法通过用不同浓度的香加皮杠柳苷对SP2/0小鼠骨髓瘤细胞及小鼠体内移植瘤进行干预处理,MTT法检测CPP处理的SP2/0小鼠骨髓瘤细胞的增殖抑制率,流式细胞仪检测小鼠骨髓瘤细胞的凋亡率,Westernblot检测小鼠骨髓瘤细胞及移植瘤中IL-6和DKK1蛋白的表达水平.结果不同浓度CPP能抑制SP2/0小鼠骨髓瘤细胞增殖,加速SP2/0小鼠骨髓瘤细胞凋亡,在裸鼠体内,不同浓度CPP对抑制骨髓瘤移植成瘤,同时降低骨髓瘤细胞和移植瘤中IL-6和DDK1蛋白的表达水平.结论香加皮杠柳苷能够通过抑制IL-6和DDK1表达,阻断Wnt信号来达到治疗小鼠骨髓瘤的作用,有望成为治疗多发性骨髓瘤及骨髓瘤骨病的新药物.
Abstract:
Objective To investigate the effects and mechanism of action of the periplocin from cortex periplocae(CPP) on proliferation,apoptosis and transplantation of myeloma cells in mice.Methods The different concentrations of the periplocin from cortex periplocae were used on SP2/0 myeloma cells and mice transplanted tumor.The proliferation inhibition rate of SP2/0 myeloma cells was determined by MTT method,the cells apoptosis rate of SP2/0 myeloma cells was determined by flow cytometer,the expression level of IL-6 and DKK1 in SP2/0 myeloma cells and mice transplanted tumor were analyzed by Western blot.Results The proliferation of SP2/0 myeloma cell was inhibited and the apoptosis of SP2/0 myeloma cell was accelerated by different concentrations of CPP,the myeloma transplanted tumor was inhibited and the expression level of IL-6 and DDK1 were reduced by different concentrations of CPP myeloma cells in SP2/0 myeloma cell and transplanted tumor.Conclusion CPP could inhibit IL-6 and DDK1 expression,and block Wnt signal to achieve the effect of treatment of mice myeloma.It is expected to be a new drug for the treatment of multiple myeloma(MM) and myeloma bone disease(MBD).
