摘要:
目的 探讨非染色体结构维持蛋白复合体I G亚基(Non-SMC condensin I complex subunit G,NCAPG)对大肠癌细胞生长的作用和对增殖凋亡的影响。方法 敲减NCAPG的小干扰RNA(si-NCAPG)转染人大肠癌细胞株HCT116,Western blot检测si-NCAPG对大肠癌细胞中NCAPG蛋白表达的影响; 通过观察细胞形态、细胞活力评价si-NCAPG对大肠癌细胞生长的影响,并筛选效果最好的序列用于后续实验。通过克隆形成、细胞周期和凋亡、Western blot实验评价si-NCAPG对大肠癌细胞生长、周期和凋亡的影响。结果 si-NCAPG显著降低大肠癌细胞中NCAPG的蛋白表达(P<0.05),抑制大肠癌细胞数目和细胞活力(P<0.05),使细胞阻滞在G2/M期(P<0.05),并促进细胞凋亡(P<0.05)。敲减NCAPG降低Cyclin B1、CDK1和Bcl-2蛋白表达水平和升高BAX的蛋白表达水平(P<0.05)。结论 敲减NCAPG通过阻滞细胞在G2/M期和促进细胞凋亡,达到抑制大肠癌细胞生长的作用。
Abstract:
Objective To investigate the effect and mechanism of Non-SMC condensin I complex subunit G(NCAPG)on the growth,proliferation and apoptosis of colorectal cancer cells(CRC).Methods siRNA-NCAPG was transfected into human colorectal cancer cell line HCT116 cells.The effect of si-NCAPG on the expression of NCAPG in HCT116 cells was detected by Western blot.The effect of si-NCAPG on the growth of HCT116 cells was evaluated by observing cell morphology and cell viability, and the best sequences were screened for subsequent experiments.The effects of si-NCAPG on the cell growth, cycle and apoptosis of HCT116 cells were evaluated by clonal formation, cell cycle, apoptosis, and western blot.Results si-NCAPG significantly decreased the expression of NCAPG protein level(P<0.05), inhibited the cell number and cell viability(P<0.05), induced cell cycle arrest in G2/M phase(P<0.05)and promoted cell apoptosis in HCT116 cells(P<0.05).Knocking down NCAPG reduced expression levels of Cyclin B1, CDK1 and Bcl-2 protein and increased expression levels of BAX protein.Conclusion si-NCAPG significantly inhibits the growth of CRC by inducing cell cycle arrest at G2/M phase and promoting cell apoptosis.
