摘要:
目的 探讨circ_0089153对TPC-1甲状腺癌细胞增殖、迁移及侵袭的影响。方法 采用circ_0089153 shRNA沉默TPC-1细胞中的circ_0089153表达并分成空白对照组、阴性对照组及shRNA沉默组,通过RT-PCR及Western blot检测不同组TPC-1细胞中circ_0089153 mRNA及蛋白的表达,采用CCK-8法检测不同组TPC-1细胞增殖,采用Transwell实验检测不同组细胞迁移和侵袭能力。结果 shRNA沉默组circ_0089153 mRNA及蛋白表达水平较空白对照组、阴性对照组低(P<0.05); 沉默hsa_circ_0089153后,TPC-1细胞增殖受到抑制; shRNA沉默组细胞增殖率、迁移和侵袭的细胞数较对照组低(P<0.05)。结论 circ_0089153与甲状腺癌细胞增殖、迁移及侵袭密切相关。
Abstract:
Objective To investigate the effects of circ_0089153 on the proliferation, migration and invasion of TPC-1 thyroid cancer cells.Methods The expression of circ_0089153 in TPC-1 cells was silenced by circ_0089153 shRNA and divi-ded into blank control group, negative control group and shRNA silenced group.The expression of circ_0089153 mRNA and protein in TPC-1 cells in different groups was detected by RT-PCR and Western blot, the proliferation of TPC-1 cells in different groups was detected by CCK-8 method, and the migration and invasion ability of TPC-1 cells in different groups was detected by transwell assay.Results The mRNA and protein expression levels of circ_0089153 in shRNA silencing group were lower than those in blank control group and negative control group(P<0.05).After silencing hsa_circ_0089153, TPC-1 cell proliferation was inhibited.The number of cell proliferation, migration and invasion in shRNA silencing group was lower than that in control group(P<0.05).Conclusion circ_0089153 is closely related to the proliferation, migration and invasion of thyroid cancer cells.
