摘要:
目的 研究中性粒细胞源性外泌体对肺腺癌细胞迁移和侵袭的影响。方法 采用1 μmol/L全反式维甲酸(ATRA)诱导人早幼粒细胞白血病NB4细胞分化成中性粒细胞,MGG染色分析其形态学的改变,流式细胞技术检测其表面标志物CD11b及CD18的表达。采用总外泌体试剂盒分离提取NB4细胞和中性粒细胞来源的外泌体,利用透射电镜技术鉴定外泌体的形态与大小。通过细胞划痕和Transwell迁移实验分析NB4细胞和中性粒细胞来源的外泌体对A549细胞迁移能力的影响。采用Transwell侵袭实验研究NB4细胞和中性粒细胞来源的外泌体对A549细胞侵袭能力的影响。结果 MGG染色结果显示NB4细胞经ATRA诱导后分化成中性粒细胞,且流式细胞检测结果表明中性粒细胞表面标志物CD11b及CD18表达明显升高。电镜下观察到NB4细胞和中性粒细胞来源的外泌体大小为40~100 nm,形态呈双层膜,中空,圆形或椭圆形。细胞划痕和Transwell实验证实,中性粒细胞源性外泌体比NB4细胞源性外泌体能更明显地增强肺腺癌细胞的迁移和侵袭能力。结论 中性粒细胞源性外泌体能明显促进肺腺癌细胞的迁移和侵袭,参与肺腺癌的发生发展。
Abstract:
Objective To investigate the influence of neutrophil-derived exosomes on migration and invasion of lung adenocarcinoma cells.Methods All-trans retinoic acid(ATRA)was used to induce differentiation of human promyelocytic leukemia NB4 cells into neutrophils.The morphologic changes of neutrophils were analyzed by MGG staining, and the expressions of CD11b and CD18 for the neutrophils specific markers were detected by flow cytometry.The total exosome kit was used to isolate and extract exosomes derived from NB4 cells and neutrophils, the morphology and size of exosomes were identified by transmission electron microscopy.The wound healing and Transwell assay were used to investigate the effects of NB4-derived and neutrophil-derived exosomes on the migration and invasion of A549 cells.Results The NB4 cells were differentiated into neutrophils successfully and the expressions of CD11b and CD18 were significantly increased after ATRA induction.The NB4-derived and neutrophil-derived exosomes ranged from 40 nm to 100 nm, and were bilayer, hollow, round or oval.The NB4-derived and neutrophil-derived exosomes could promote the migration and invasion of A549 cells, and the effect of neutrophil-derived exosomes in promoting the migration and invasion of A549 cells was more pronounced.Conclusion Neutrophil-derived exosomes could promote the migration and invasion of lung adenocarcinoma cells significantly, and play a major role in the occurrence and progression of lung adenocarcinoma.
